Hybridization Denaturing Solution
This denaturing solution is a strong base that denatures double-stranded DNA so that it can be efficiently transferred onto a nylon membrane and subsequently be hybridized to a labeled probe. When used with a depurination solution, the treatment results in the hydrolysis of the phosphodiester backbone at the sites of depurination, allowing for more efficient transfer of large DNA in neutral Southern transfers.
The denaturing solution is composed of 0.5N NaOH, and 1.5M NaCl
