The Acid-Fast (Ziehl-Neelsen) Stain is a widely used diagnostic staining method for detecting acid-fast organisms, most notably Mycobacterium species such as M. tuberculosis, the causative agent of tuberculosis. This staining technique relies on the unique ability of certain bacteria to retain the primary stain (carbol fuchsin) even when exposed to decolorization with acid-alcohol. The counterstaining with methylene blue allows for the clear visualization of these acid-fast bacteria, which appear red against a blue background, facilitating their identification under a microscope.
